Aim : Standard plate count method to check viable cells
Principle: There are microbiology techniques which helps us to count and check viability of microbes present in sample.sample are collected from air , water and soil .
These methods are :
Spread plate method : this method uses a L shaped loop made up of glass or metal with smooth surface for spreading microbes from sample over a plate which tells us about number of microbial colony in sample from which inoculation was taken .
Pour plate : This technique uses molten nutrient agar for growth of mixed culture in the agar . Heat sensitivity bacterium growth are reduced in this method as agar is hot and can effect growth of heat sensitive and obligate anaerobes.
Sterak plate methods:This technique involves T-steak and Quadrant method
This method of bacterial culture involves continuous striking to get separated colonies of microbes from mixed samples . Individual colonies are isolated in this technique.
Observation:different platting methods are used to isolate colonies from mixed culture and used to check viability of colonies microbes.colonies of pure culture are isolated.
Result:different pure colonies were isolated and vibility can be checked .
