RFLP


Restriction fragment length polymorphism .this is a non PCR based technique invented by Alec Jeffreys during his research in hereditary diseases .this technique is used to analysis different patterns in DNA  mapping of different organisms. The recognised by RFLP are called variable number tandem repeats (VNTPs). In this method ,DNA is digested by restriction enzymes which cut the DNA at specific sequences ,electrophoresed ,blotted on a membrane and probed with a ladled clone. The DNA sequence variations detected by this method were called as RFLP.


Procedure


Some basic steps are followed in this of performing RFLP technique:


 DNA isolation 


 Using restriction enzymes to cut DNA into smaller fragments.


 Separation of DNA by get electrophoresis .


 Transfer of DNA fragments onto a nylon or nitrocellulose membranes filter


 Using labelled probe visualisation of DNA  obtained 


 Analysis of results .


 Genomic DNA  of an organism is isolated and digested with a particular restriction endonuclease .
